COMPOSITION
100 ml of the product contains active substances, g:
Glucose monohydrate – 37.40; L-cysteine – 0.66, Sodium citrate – 4.95, Sodium hydrogen carbonate – 1.38, Trilon B – 1.38, Potassium chloride – 0.67, Citric acid – 0.22, Colistin sulfate – 0.28, Gentamycin sulfate – 0.28, water for injection up to 100 ml.
PHARMACOLOGICAL PROPERTIES
4D is a diluent for boar semen and its short-term storage.
4D provides optimal conditions for preserving the biological usefulness of spermatozoa and their high fertility, maintains the integrity of the membrane apparatus and intracellular subunits, which provides the high functional activity of the sex cells.
Gentamicin sulfate and colistin sulfate as parts of the diluent provide an antimicrobial effect on the microflora that may be contained in the semen of boars.
4D causes slowing of metabolism, thereby increasing the spermatozoa physiological resistance to thermal shock, prevents agglutination and prolongs their survival outside the body of the male.
pH of working solution at 20°C is 6.6-7.4; osmolarity is 300-350 mOsm/L.
USE OF THE DILUENT
Diluent solution (concentrated) is designed for dilution of boar semen with subsequent its storage in conditions in vitro at a temperature of 17 ± 1°C and insemination of sows.
DOSAGE
To prepare the diluent working solution for the boar semen, the content of the vial is poured into a container with a corresponding amount of bidistilled water (according to Table 1). The solution is stirred by slow rotational movements to achieve uniformity.
Before semen dilution the finished diluent working solution should be kept for 10-15 minutes at a temperature of 30-35°C in a water bath (thermostat) or on a heating plate (to balance the physical and chemical properties of the solution).
Diluent working solution maintains its properties for 48 hours when stored in a refrigerator at a temperature between 1 and 5°C in a hermetically sealed container.

Freshly obtained boar semen before its dilution should be stored in a thermo container at a temperature of 30-35 ° C. Before semen dilution, its parameters: volume, concentration and motility of spermatozoa should be determined. Ratio of dilution of boar semen depends on the concentration of spermatozoa in 1 ml and their motility. Allowable ratio of dilution of boar semen is 1: 3 to 1:10.
Dilution of semen is carried out not earlier than 10 minutes and not later than 20 minutes after its collection. Before dilution of native semen, one must control the temperatures of ejaculate and diluent. During the process of dilution, temperature of the semen and diluent should be the same (maximum permitted deviation ± 1°C). Ready diluent by small portions slowly is added to semen (and not vice versa!). Then the diluted semen should be stirred by careful circular motions.
After dilution of the semen, it is packed into vials. Semen packaging is carried out at room temperature 20-22°C. Ready semen doses are kept at room temperature for 1.5-2 hours, protecting them from light and hypothermia. Farther, the vials with semen are placed in the Climabox at a temperature of 17 ± 1°C and stored in a horizontal position. Semen doses, which are stored in Climabox for several days, should be turned over daily.
Diluted semen is suitable for artificial insemination of sows within 4 days from the time of its dilution, provided that it is properly stored.
IT IS NOT NECESSARY TO PREHEAT SPERMODOSES BEFORE ARTIFICIAL INSEMINATION OF SOWS!
